چکیده

A cDNA clone for NADPH : protochlorophyllide oxi doreductase from barley was subcloned for production of antisense-mRNA. This enabled heterologous hybridi zation with RNA from cress seedlings (Lepidium sativum L.). The mRNA level for N A DPH : protochlorophyllide oxidoreductase did not decrease in cress seedlings during irradiation with continuous far-red or white light up to 12 h. The amount of NADPH : protochlorophyllide oxi doreductase protein, identified by Western blot de creased 5-fold after continuous irradiation with white light for 12 h. Species differences for light regulation of RNA are discussed.

Enzymic Properties of Purified Myrosinase from Lepidium sativum Seedlings

To establish the substrate specificity o f the thioglucoside glucohydrolase myrosinase (EC 3.2.3.1), this enzyme was purified to homogeneity from light-grown cress (Lepidium sativum L.) seedlings by Sephadex gel filtration. Red Dye and anion exchange (FPLC M ono Q) chro matography, and preparative isoelectric focusing. Hydrolytic activity was shown toward only 4 o f the 29 synthetic and natura...

Glucosinolate Biosynthesis: Sulfation of Desulfobenzylglucosinolate by Cell-Free Extracts of Cress (Lepidium sativum L.) Seedlings.

After removal of myrosinase activity by concanavalin A-Sepharose 4B chromatography, cell-free extracts of light-grown cress (Lepidium sativum L.) seedlings, catalyzed the sulfation of desulfobenzylglucosinolate (K(m), 0.23 millimolar) to benzylglucosinolate using PAPS (K(m), 1 millimolar) as sulfur donor. Sulfotransferase activity, which was optimal at pH 9.0, was stimulated by MgCl(2), MnCl(2)...

Potent endogenous allelopathic compounds in Lepidium sativum seed exudate: effects on epidermal cell growth in Amaranthus caudatus seedlings

Many plants exude allelochemicals--compounds that affect the growth of neighbouring plants. This study reports further studies of the reported effect of cress (Lepidium sativum) seed(ling) exudates on seedling growth in Amaranthus caudatus and Lactuca sativa. In the presence of live cress seedlings, both species grew longer hypocotyls and shorter roots than cress-free controls. The effects of c...

Development of a Biocrystallisation Assay for Examining Effects of Homeopathic Preparations Using Cress Seedlings

A major challenge in basic research into homeopathic potentisation is to develop bioassays that yield consistent results. We evaluated the potential of a seedling-biocrystallisation method. Cress seeds (Lepidium sativum L.) germinated and grew for 4 days in vitro in Stannum metallicum 30x or water 30x in blinded and randomized assignment. 15 experiments were performed at two laboratories. CuCl...
